Anti?inflammatory activities of Gardenia jasminoides extracts in retinal pigment epithelial cells and zebrafish embryos

چکیده

Age?related macular degeneration (AMD) is the most common cause of visual impairment in developed countries. Inflammation serves a critical role pathogenesis AMD. Gardenia jasminoides found several regions China and traditionally used as an organic yellow dye but has also been widely therapeutic agent numerous diseases, including inflammation, depression, hepatic vascular disorders, which may reflect variability functional compounds that are present extracts (GJE). To investigate potential GJE for AMD, ARPE?19 cells were treated with lipopolysaccharide (LPS) or LPS plus GJE. significantly decreased LPS?induced expression proinflammatory cytokines, IL?1?, IL?6 TNF??. In in vivo study, inhibited CuSO4?induced migration primitive macrophages to lateral line zebrafish embryos. attenuated cytokines (IL?1?, TNF??), NFKB activating protein (nkap) TLR4 cells. The results study demonstrated anti?inflammatory vitro and vivo, suggested candidate

Extremely low frequency-pulsed electromagnetic fields affect proangiogenic-related gene expression in retinal pigment epithelial cells

Objective(s): It is known that extremely low frequency-pulsed electromagnetic fields (ELF-PEMF) influence multiple cellular and molecular processes. Retinal pigment epithelial (RPE) cells have a significant part in the emergence and pathophysiology of several ocular disorders, such as neovascularization. This study assessed the impact of ELF-PEMF on the proangiogenic features of RPE cells. Mate...

Actin-dependent pigment granule transport in retinal pigment epithelial cells.

Since diffusion of particles as large as vesicles or organelles is constrained by the high viscosity of cytoplasm, directed translocation of these particles from one cytoplasmic site to another must be achieved by cytoskeletal intervention. In most cases, this is achieved by attaching the particle to a molecular motor which then navigates along a microtubule or an actin filament track (Fath and...

Uptake by Retinal Pigment Epithelial Cells

Uptake of Ca was studied in isolated frog retinal pigment epithelial cells. Ca accumulation was found to be a saturable, temperature-dependent event. Kinetic analysis of this accumulation revealed two transport systems with apparent km of 2.0 and 0.3 mM. We found the presence of a Na-Ca exchanger mechanism that releases Ca under depolarized conditions. Light induced an increase of Ca uptake due...

osteoclast-like cells in early zebrafish embryos

objective: genes involved in bone and tissue remodelling in the vertebrates include matrix metalloproteinase-9 (mmp-9), receptor activator of necrosis factor κ-β (rank), cathepsin-k (ctsk) and tartrate-resistant acid phosphatase (tracp). we examine whether these markers are expressed in cells of zebrafish embryos of 1-5 days post fertilization. we also examine adult scales, which are known to c...

Phagocytosis and surface morphology in cultured retinal pigment epithelial cells.

Phagocytosis of latex spheres was studied in explant cultures of retinal pigment epithelium from rabbit, calf, and rat (normal and dystrophic). Calf and rabbit pigment epithelial cells showed a "latent period" of about 12 and 17 hr, respectively, during which latex spheres were not ingested even when the cultures were pre-incubated for up to 24 hr prior to exposure. Scanning electron microscopy...
